Quality Assurance: Ensuring Authentic Nefiracetam Powder

Nefiracetam is a cognitive enhancer or nootropic that helps combat dementia or Alzheimer’s by regulating the brain’s GABA (gamma-aminobutyric acid) levels. It also has affinity for the cholinergic and monoaminergic systems which help boost learning, focus, and memory.

Evaporative crystallization experiments in ethyl acetate at room temperature led to the formation of an ionic cocrystal with zinc chloride and one water molecule, named NZCW.

Molecular Structure

Nefiracetam powder europe does not occur in nature and is exclusively synthesized in laboratories. It is a fat soluble racetam molecule and shares structural similarity with Piracetam and Aniracetam, which are also cognitive enhancement compounds that are not found naturally.

Nefiracetam is a poorly soluble compound with low water solubility (BCS class II). It exhibits deliquescence in hygroscopic organic solvents, and its oral absorption and bioavailability are consequently low15. Therefore, formulation tools must be applied to increase its dissolution rate and bioavailability17.

In XRPD experiments, the monohydrate of nefiracetam (FI) showed a transition to a second polymorph nefiracetam (FII) at temperatures around 135-145 degC. Upon heating, the FII reverted back to the FI form.

The simulated XRPD patterns of FI, FII, NCA, and NOA cocrystals clearly distinguish their structures. Table 3 shows the corresponding dissolution rates and solubility at 18 degC in MeCN for these four forms. Their congruent solubility is similar to that of the pure FI.

Biological Activity

Nefiracetam is a nootropic drug of the racetam family and is also known as DM-9384. It helps improve cognitive ability, memory, focus, and concentration. It works by enhancing the brain’s GABA (gamma-aminobutyric acid) activity. This is the primary inhibitory neurotransmitter in your brain and it keeps glutamate levels from getting too high, which damages the brain. It also increases cholinergic and monoaminergic systems, both of which are important for learning and memory.

It prolongs the opening of calcium channels, keeping neurotransmitters active longer. It also potentiates acetylcholine receptors, which boosts a calming effect. Nefiracetam has also shown to be neuroprotective in animal models of ischemic brain damage.

Several anhydrous polymorphs of nefiracetam have been identified by screening for physicochemical properties using a range of solvents including ethyl acetate, acetone, and dimethyl sulfoxide. A cocrystallization study using the biocompatible coformers citric acid, oxalic acid, and zinc chloride led to the identification of 13 novel cocrystals that have significantly improved thermal stability, hygroscopicity, solubility, and dissolution rates compared with those of the parent compound.

Pharmacology

Nefiracetam powder (DM-9384) is a member of the racetam family. Structurally, it resembles piracetam and aniracetam. Like all members of the racetam family, nefiracetam is a fat-soluble compound that easily crosses the blood-brain barrier and works in the brain by enhancing GABAergic, cholinergic, and monoaminergic neuronal systems. It has been shown to reduce apathy, improve cognition and memory formation, and promote healthy brain cells.

It also enhances the activity of nicotinic acetylcholine receptors, which increases the release of both GABA and glutamate. This boosts neuroplasticity and learning, while providing a calming effect and reducing symptoms of anxiety and depression.

Nefiracetam FI, FII and monohydrate were characterized by single-crystal X-ray diffraction using a Cooling Thermomixer HLC and data measured on a Rigaku Ultra X18S generator with FOX3D mirrors. The slurry crystals were prepared by slurrying the compound overnight in ethyl acetate and then allowing the solvent to evaporate, leaving the solid phase. Slurry crystals were retrieved and dried.

Dosage

Like most racetam compounds, nefiracetam takes longer than piracetam to show results and needs prolonged supplementation in order to maximize memory formation. It also requires a good stack with choline, such as CDP-Choline or Alpha GPC to boost neural acetylcholine.

Nefiracetam is considered non-toxic and well-tolerated at recommended doses. Some side effects that occur rarely include headaches, anxiety and fatigue. These are usually a result of taking too high of a dosage.

Neurohackers use nefiracetam to boost their learning and memory, especially long-term memory. It helps boost the activity of GABA, glutamate and acetylcholine in the brain. It is also known to increase attention span and improve cognitive function. Early research suggests that it may have neuroprotective properties and help prevent cognitive decline caused by stroke. Nefiracetam is a good choice for students and executives looking to improve their study scores, work flow and concentration. It’s also been shown to improve apathy and depression in patients with cerebrovascular disease.
